Title : Development of Acoustic Force Spectroscopy to study single molecule unbinding and single cell viscoelasticity
Abstract:
Adhesion and mechanics are interdependent and it is difficult to understand one without the other. Their experimental characterisation involves applying forces and measuring the resulting displacement or deformation. This can be done using Acoustic Force Spectroscopy (AFS), a new tool that uses acoustic waves to manipulate beads or cells in a microfluidic channel.
At the single molecule level, we are using this technique to probe the unbinding kinetics of receptor-ligand complexes using cyclic force measurements, and to study the near-equilibrium unbinding of the model streptavidin-biotin complex.
At the cellular level, we are coupling it with Reflection Interference Contrast Microscopy (RICM) to measure the mechanics of suspended cells, without contact or labelling.
These two applications have required the development of new calibration methods and allow to access slow regimes (at low loading rates and low frequencies) that are still relatively unexplored.
